Lustre started as a TV host in RPN's children oriented show entitled Storyland, when she was nine years old. She became one of the members of the cast in TV5's youth-oriented program Bagets, being a talent of Viva Entertainment. Lustre also appeared in the film Petrang Kabayo with Vice Ganda on the lead role, and starred in P. S. I Love You a drama series in TV5.

Lustre appeared in several GMA Network shows including performances in SOP Rules.

When Viva Films got the film rights of the best-selling novel Diary ng Panget by HaveYouSeenThisGirL, she got the role of the main protagonist, Eya Rodriguez. She starred together with James Reid, Yassi Pressman and Andre Paras. The film was released on April 2, 2014 and became a box-office hit earning at least 120 million pesos during its 4-week run.

Lustre together with James Reid and Joseph Marco, starred in another film adaptation entitled Talk Back and You're Dead, which is based on the best-selling book of the same name by Alesana Marie. The film was released on August 20, 2014, in theaters nationwide.

On August 26, 2014, Lustre along with on-screen partner Reid signed a two-year exclusive contract with ABS-CBN (as ABS-CBN reacquired Reid) that was attended by some of the prominent figures of the station. For her television projects with Reid on ABS-CBN, she portrayed the lead female role Anika Andres in the Wansapanataym month-long special entitled My App Boyfie in October, and Carmina Joven in their MMK debut on its Valentine's Day episode.

She starred again with Reid in Para sa Hopeless Romantic, a film adaptation of the best-selling novel with the same name written by Marcelo Santos III. The movie was released on May 13, 2015. She also had a special role in the horror film Chain Mail by Viva Films, which premiered on August 22, 2015. Among Google's top ten most search celebrity in the Philippines, she is ranked second in the female category.

She currently stars as Leah Olivar on the ABS-CBN Primetime Bida series On The Wings Of Love, starring alongside Reid as her leading man.

MARAMING Pinoy ang nalungkot nang tawagin ng host ng Miss Universe 2015 na si Steve Harvey ang pangalan ng Miss Colombia bilang Miss Universe 2015, bale ang pambato nating si Pia Wurztbach ang lumalabas na 1st Runner-Up kaya pumunta na siya sa likod samantalang si Miss Colombia Ariadna Gutierrez ay tuwang-tuwang kinoronahan agad ng reigning Miss Universe 2014. also from Colombia, na si Paulina Vega.

Feeling ko tuloy ang swerte-swerte naman ng Miss Colombia kasi back-to-back win din sila tulad ng Philippines sa Miss Earth dahil pareho nating nakuha ang korona for  Miss Earth 2014 and Miss Earth 2015.

Sa loob-loob ng mga kababayan natin, e, talaga yatang forever runner-up na lang tayo sa Miss U.  Nakapanlulumo.  Pero maya-maya ay bumalik sa stage ang host ng Miss U na si Steve Harvey na nagso-sorry sa audience at sa milyun-milyong televiewers around the globe na siya ay nagkamali dahil ang totoo raw nanalong Miss Universe 2015 ay si Miss Philippines.  

Nagulat si Pia.  Natulala naman si Miss Colombia.   Halos hindi na siya makagalaw at makangiti sa sobrang pagkapahiya.  Naguluhan ang audience.  Nabuhayan naman ng loob ang mga Pinoy na personal na nanood sa Planet Hollywood sa Las Vegas kung saan ginanap ang prestihiyosong beuaty pageant.

Sa puntong yun ay panay naman ang sorry ng nagkamaling host. Aniya, yun daw talaga ang nakalagay sa card na binasa niya, si Miss Colombia ang 1st runner-up at si Miss Philippines ang Miss Universe 2015. Sabi pa niya, 'Please dont blame the ladies.  I'll take full responsibility for my mistake,' or something to that effect.






 

Sa totoo lang, tuwing ipinapakita sa TV screen ang scores ng mga candidates for each round competition like sa swimsuit (Top 15), evening gown (Top 10) at question and answer portion (Top 5) ay palaging nangunguna ang Philippines sa Online votes at palagi namang pumapangalaawa sa kanya ang Miss Colombia. So, parang ang online votes ay sumasalamin sa boto ng limang judges dahil sa huli ay silang dalawa pa rin (Miss Philippines and Miss Colombia) ang naging mahigpit na magkalaban.
